How long does it take the bus from Kailua to downtown during mon-fri 7am'ish? and then again after work from downtown back to kailua around 6pm?

what about by car?

what about by car?

About an hour each way at those times by car. You're going with rush-hour traffic.

Much less at other times of the day. I work nights and leave my house off Keolu Drive at about 6:10 in the evening or so and arrive at Punchbowl Street at about 6:35, give or take a few minutes.
